Biological Activity
INH14 is a small-molecule inhibitor targeting TLR2-mediated NF-κB activation through selective action on IKKα and IKKβ. By blocking the catalytic subunits of the IκB kinase complex, INH14 inhibits IκBα phosphorylation and subsequent nuclear translocation of the p65/p50 NF-κB dimer. This signaling pathway disruption downregulates TLR2-driven pro-inflammatory cascades, leading to reduced expression of downstream inflammatory cytokines and cell survival phenotypes. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
