INF39 is a nontoxic, irreversible NLRP3 inhibitor that directly interacts with NLRP3 to block NLRP3 inflammasome assembly and activation. By directly targeting NLRP3, INF39 inhibits caspase-1 activation and prevents the maturation and secretion of pro-inflammatory cytokines IL-1β and IL-18. Furthermore, INF39 partially suppresses LPS-driven pro-inflammatory gene expression, thereby reducing cellular inflammatory cascades and pyroptotic cell death downstream of NLRP3 activation. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
