Tudociclib (INCB123667) is a potent and selective small-molecule inhibitor of cyclin-dependent kinase 2 (CDK2). By targeting CDK2, it disrupts the activation of cyclin E-CDK2 and cyclin A-CDK2 complexes, thereby blocking phosphorylation of retinoblastoma protein (Rb) and inhibiting E2F-mediated gene transcription. Consequently, tudociclib arrests cells at the G1/S phase boundary of the cell cycle and suppresses tumor growth in CDK2-dependent cancer models. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
