Biological Activity

INCA-6 is a cell-permeable inhibitor of nuclear factor of activated T-cells (NFAT) signaling. It selectively blocks the docking of phosphorylated NFAT to calcineurin, thereby preventing calcineurin-dependent NFAT dephosphorylation and downstream calcineurin-NFAT signaling. By impairing calcineurin-NFAT complex formation, INCA-6 selectively suppresses NFAT-mediated gene transcription and downstream cellular responses without inhibiting calcineurin phosphatase activity toward other substrates. FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
