IID432 is a potent oral cyanotriazole inhibitor of Trypanosoma cruzi topoisomerase II, exhibiting an EC50 of 8 nM. By selectively targeting topoisomerase II, IID432 disrupts parasite DNA replication and genomic maintenance. This inhibition leads to complete washout-resistant parasite clearance in vitro and promotes rapid in vivo clearance of Trypanosoma cruzi infections in animal models, demonstrating strong potential for short-course Chagas disease therapy.
