Biological Activity

Iclepertin (BI-425809) is a potent and selective small-molecule inhibitor of glycine transporter 1 (GlyT1). By inhibiting GlyT1-mediated glycine reuptake, it elevates extracellular glycine levels in the central nervous system and cerebrospinal fluid. This increase promotes N-methyl-D-aspartate (NMDA) receptor activity via enhanced co-agonist site occupancy, facilitating downstream long-term potentiation, synaptic plasticity, and cognitive function. FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
