Ordering Information

Biological Activity

IBG1 is an intramolecular bivalent molecular glue degrader targeting BET bromodomains, promoting proteasomal degradation of BRD4 with a DCof. By bivalently engaging adjacent bromodomains of BRD4 in cis, IBG1 enhances surface complementarity and recruits the CRL4-DCAF16 E3 ubiquitin ligase complex. This induced proximity triggers BRD4 ubiquitination and subsequent degradation, leading to downstream inhibition of cancer cell proliferation. [1]

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
