Biological Activity

IAXO-102 is a selective small-molecule TLR4 antagonist that directly targets both MD-2 and CD14 accessory co-receptors. By interfering with TLR4 coreceptor signaling complexes, it inhibits downstream MAPK and p65 NF-κB phosphorylation. This pathway inhibition downregulates the expression of TLR4-dependent proinflammatory proteins, attenuates inflammatory cellular responses, and suppresses the development of experimental abdominal aortic aneurysm. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
