IAM1363 is a selective, irreversible, and brain-penetrant HER2 inhibitor. It targets the tyrosine kinase domain to block autophosphorylation and activation of both wild-type HER2 and activating HER2 mutant variants. By disabling HER2 kinase activity, IAM1363 suppresses downstream oncogenic cascades including the MAPK and AKT/mTOR signaling pathways, leading to the inhibition of cellular proliferation and survival in HER2-driven tumor cells. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
