Biological Activity

I-138 is an orally active, potent reversible inhibitor of ubiquitin-specific protease 1 (USP1)-UAF1 with an IC50 of 4.1 nM. Structurally related to ML, the compound increases FANCD2 and PCNA monoubiquitination in cells, disrupting cellular DNA repair mechanisms. Downstream, USP1 inhibition leads to decreased DNA synthesis, replication stress, S-phase arrest, and accumulation of DNA damage, selectively inducing synthetic lethality in BRCA1/2-mutant cancer cells. FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
