HTH-01-015 is a potent and selective kinase inhibitor targeting NUAK1 (IC50 100 nM) with >100-fold selectivity over NUAK2. By inhibiting NUAK1 activity, this compound disrupts downstream phosphorylation cascades involving substrate proteins such as MYPT1. Attenuation of NUAK1 signaling alters cellular motility, suppresses cell proliferation, and regulates cytoskeletal dynamics in human cancer cell models.

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
