HSP27 inhibitor J2 is a small-molecule inhibitor targeting heat shock protein 27 (HSP27). It induces abnormal HSP27 dimer formation and suppresses the assembly of giant HSP27 polymers, thereby disrupting its protein chaperone activity and cellular cytoprotective function. Downstream, J2 sensitizes cancer cells to chemotherapeutic agents, enhancing the antiproliferative activity of 17-AAG and increasing cisplatin-induced growth inhibition in lung cancer cells. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
