Biological Activity

HS-276 is a potent, orally bioavailable, and highly selective transforming growth factor-β-activated kinase 1 (TAK1) inhibitor with a Ki of 2.5 nM. Inhibition of TAK1 disrupts TNF-mediated intracellular signaling and downstream NF-κB activation, thereby reducing proinflammatory cytokine profiles. Consequently, HS-276 suppresses inflammatory responses, leading to significant attenuation of disease severity and arthritic-like symptoms in collagen-induced arthritis mouse models. [1]

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
