Biological Activity

HS-1371 is an ATP-competitive inhibitor of receptor-interacting protein kinase 3 (RIP3). By occupying the ATP-binding site of RIP3, the compound blocks RIP3 enzymatic activity and inhibits downstream necroptotic signaling pathways. Consequently, HS-1371 suppresses TNF-induced programmed necrosis and protects cells from RIP3-dependent necroptosis, serving as a valuable chemical tool for investigating necroptosis-mediated cellular injury and inflammatory cascades. FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
