Biological Activity

HRX215 (Darizmetinib) is a first-in-class, selective, orally active mitogen-activated protein kinase kinase 4 (MKK4) inhibitor with an ICof. By suppressing MKK4 within the stress-activated protein kinase pathway, HRX215 redirects signaling through MKKand JNK1 to activate pro-regenerative gene programs. This targeted inhibition promotes hepatocyte proliferation, accelerates liver tissue regeneration, and prevents liver failure in preclinical models. [1]

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
