HRO761 is a potent, selective, allosteric inhibitor of Werner syndrome RecQ helicase (WRN). By binding at the interface of the D1 and D2 helicase domains, HRO761 locks WRN in an inactive conformation and promotes target degradation. This inhibition impairs genomic stability maintenance, triggering DNA damage accumulation and selective tumor cell growth inhibition in microsatellite instability (MSI) cancer cells. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
