Biological Activity

HOIPIN-8 is a potent inhibitor of the linear ubiquitin chain assembly complex (LUBAC, IC50 = 11 nM) that suppresses NF-κB signaling [1]. By blocking LUBAC-mediated linear ubiquitination, it downregulates TNF-α-stimulated canonical NF-κB activation and pathogen-induced immune pathways [1, 2]. Consequently, HOIPIN-8 impairs downstream inflammatory cytokine expression, alters cell death responses, and serves as a valuable tool for investigating LUBAC-dependent physiological functions in cellular immunity.[1][2]

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
