Biological Activity

Hoechst 33342 is a cell-permeable fluorescent nucleic acid stain that specifically binds to adenine-thymine-rich regions in the minor groove of double-stranded DNA. Upon minor groove binding, the dye exhibits enhanced fluorescence quantum yield, enabling clear visualization of nuclear morphology and chromatin organization in both interphase and mitotic cells. This selective DNA association facilitates live-cell nuclear imaging, cell cycle analysis, and detection of apoptotic nuclear condensation. [1][2]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
