Biological Activity

Hexaminolevulinate hydrochloride is a lipophilic hexyl ester prodrug of 5-aminolevulinic acid that serves as a precursor for the photosensitizer protoporphyrin IX. Upon cellular entry, it increases intracellular protoporphyrin IX accumulation and, following photoactivation, triggers cytochrome c release, caspase-3 and caspase-9 activation, and apoptosis via both caspase-dependent and apoptosis-inducing factor-mediated caspase-independent pathways. Photoactivated hexaminolevulinate also reduces MDR1 P-glycoprotein expression to reverse multidrug resistance in human cancer cells. [1][2]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
