Biological Activity

Siremadlin (HDM201) is a potent and selective inhibitor of the p53-MDM2 interaction. By disrupting the binding of MDM2 to p53, Siremadlin prevents MDM2-mediated degradation of p53 and reactivates p53 signaling. This leads to the upregulation of downstream target genes, resulting in cell cycle arrest and induction of apoptosis in TP53 wild-type tumor cell lines and xenograft models. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
