H3B-120 is a selective, allosteric inhibitor of carbamoyl phosphate synthetase 1 (CPS1). By selectively inhibiting CPS1, the initial and rate-limiting enzyme in the urea cycle, it disrupts mitochondrial nitrogen metabolism and downstream pyrimidine nucleotide biosynthesis. Attenuation of CPS1 activity depletes intracellular pyrimidine pools and impairs nucleic acid synthesis, ultimately suppressing cell proliferation and exhibiting anti-tumor activity in CPS1-dependent cancer cells. - Preheat media/saline to 37°C.
