Biological Activity

Olverembatinib dimesylate (GZD824) is an orally bioavailable Bcr-Abl inhibitor targeting native Bcr-Abl and the Bcr-Abl T315I mutant. By blockading kinase activity, it inhibits Bcr-Abl autophosphorylation and suppresses downstream proliferation of leukemia cell lines expressing both wild-type and T315I mutant forms. Furthermore, olverembatinib demonstrates potent antitumor efficacy and prolongs survival in animal xenograft models of chronic myeloid leukemia. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
