GSK626616 is a potent, ATP-competitive inhibitor of dual-specificity tyrosine-phosphorylation-regulated kinase 3 (DYRK3, IC50 0.7 nM). By targeting DYRK3, it blocks the phosphorylation of PRAS40, thereby preventing stress granule dissolution and impairing mTORC1 pathway reactivation following cellular stress. In addition, DYRK3 inhibition by GSK626616 disrupts the kinase-controlled disassembly of multiple membraneless cytosolic organelles during mitosis.[1][2]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
