Biological Activity

GSK481 is a small-molecule inhibitor targeting receptor-interacting protein kinase 1 (RIP1/RIPK1). By selectively suppressing the kinase activity of RIP1, GSK481 disrupts the assembly and activation of downstream necroptotic signaling complexes. This inhibition effectively hinders necrotic cell death and blocks the release of pro-inflammatory cellular mediators, thereby modulating inflammatory cellular processes mediated by the RIP1 pathway. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
