Biological Activity

GSK269962A is a potent and selective inhibitor of Rho-associated protein kinase (ROCK), targeting ROCK1 (IC50 = 1.6 nM) and ROCK2 (IC50 = 4 nM). Inhibition of the ROCK signaling pathway reduces smooth muscle contraction and suppresses downstream phosphorylation of myosin light chain. Consequently, GSK269962A exhibits significant vasodilatory and anti-inflammatory activities, demonstrating functional efficacy in modulating vascular tone and smooth muscle relaxation. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
