Nemiralisib (GSK-2269557) is a potent and selective phosphoinositide 3-kinase delta (PI3Kδ) inhibitor with a pKi of 9.9. By targeting PI3Kδ, nemiralisib inhibits the downstream PI3K/AKT signaling pathway, suppressing leukocyte activation and pro-inflammatory cytokine production. This selective inhibition diminishes immune cell recruitment and airway inflammation, providing a therapeutic rationale for managing chronic inflammatory respiratory disorders such as asthma and chronic obstructive pulmonary disease. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
