GSK205 is a selective TRPV4 channel antagonist that inhibits TRPV4-mediated intracellular calcium influx with an IC50 of 4.19 μM. By blocking TRPV4 activation, this compound suppresses intracellular calcium signaling and inhibits downstream pathways involved in cellular mechanotransduction. Consequently, GSK205 reduces strain-induced matrix gene expression, suppresses pro-inflammatory responses, and protects against matrix degradation in articular chondrocytes.
