GSK1324726A (I-BET726) is a potent and selective inhibitor of BET family proteins, targeting BRD2, BRD3, and BRD4. By blocking the interaction of BET bromodomains with acetylated histones, it downregulates critical oncogenic driver expression, such as MYC, and suppresses pro-inflammatory cytokine production, leading to inhibition of tumor cell proliferation and anti-inflammatory phenotypes in preclinical models. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
