GSK1070916 is a potent, ATP-competitive inhibitor of Aurora B (IC50 3.5 nM) and Aurora C (IC50 6.5 nM), exhibiting over 100-fold selectivity against Aurora A. It inhibits the phosphorylation of histone H3 at serine 10, impairing mitotic progression and suppressing tumor cell proliferation. Furthermore, GSK1070916 displays dose-dependent biomarker inhibition and robust antitumor efficacy across multiple human tumor xenograft models.

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
