Biological Activity

GSK-7975A is a potent inhibitor of calcium release-activated calcium (CRAC) channels, functioning as a store-operated calcium entry (SOCE) blocker. By blocking ORAI-mediated calcium influx, it suppresses downstream calcium signaling pathways essential for immune cell activation. This compound effectively inhibits the release of mast cell mediators, including histamine and leukotriene C4, as well as pro-inflammatory cytokines such as IL-5, IL-8, IL-13, and TNF-α.[1][2]

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
