Biological Activity

GSK-is a non-covalent, selective inhibitor of DNA methyltransferase 1 (DNMT1) with an ICof. Mechanistically, it competes with hemi-methylated DNA at the active site to block DNMT1 activity, inducing proteasomal degradation of the enzyme and driving global DNA hypomethylation. Downstream, this promoter demethylation triggers transcriptional reactivation of silenced genes and inhibits cancer cell viability, demonstrating therapeutic efficacy in acute myeloid leukemia. [1]

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
