Biological Activity

GS-444217 is a potent, ATP-competitive, selective inhibitor targeting apoptosis signal-regulating kinase 1 (ASK1). By directly inhibiting ASK1 kinase activity, the compound prevents the phosphorylation and downstream activation of the JNK and p38 MAPK signaling cascades. This targeted inhibition effectively blocks oxidative stress-mediated pro-inflammatory cytokine production and suppresses cellular apoptotic responses across various cell-based experimental models. FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
