Biological Activity

GS-1427 (Emvistegrast) is a small-molecule quinolone derivative that functions as a potent and selective antagonist of α4β7 integrin. By targeting α4β7 integrin, it disrupts the interaction between α4β7 integrin and mucosal vascular addressin cell adhesion molecule-1 (MAdCAM-1). This inhibition impairs the homing of gut-tropic lymphocytes into intestinal mucosal tissues, leading to reduced downstream intestinal inflammation and therapeutic potential in inflammatory bowel disease.

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
