GRL0617 is a potent, selective, and competitive noncovalent inhibitor of severe acute respiratory syndrome coronavirus (SARS-CoV) papain-like protease (PLpro) with an IC50 ofand a Ki of. By inhibiting PLpro deubiquitinating and proteolytic activity, GRL0617 disrupts viral polyprotein processing and host immune evasion pathways, effectively blocking SARS-CoV viral replication in Vero E6 cells with an EC50 of 15 μM without causing cytotoxicity. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
