Grazoprevir is a potent Hepatitis C Virus (HCV) NS3/4A protease inhibitor targeting genotype 1a (IC50 7 pM), genotype 1b (IC50 4 pM), and genotype 4 (IC50 62 pM). By selectively inhibiting the NS3/4A protease complex, Grazoprevir blocks the cleavage of viral polyproteins required for HCV replication. This targeted enzymatic inhibition suppresses viral RNA synthesis and disrupts viral assembly in infected host cells. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
