Biological Activity

Grapiprant (CJ-023,423) is a selective prostaglandin EP4 receptor antagonist, demonstrating high binding affinity for human EP4 receptor (Ki 13 nM) and rat EP4 receptor (Ki 20 nM). It selectively inhibits PGE2-induced EP4 signaling, blocking intracellular cyclic AMP accumulation without affecting other prostanoid receptor subtypes. By targeting the EP4 pathway, grapiprant effectively mitigates pain, inflammation, and hyperalgesia in animal models of chronic inflammatory conditions. [1]

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
