Biological Activity
GNE-495 is a potent and selective inhibitor of MAP4K4. By inhibiting MAP4K4 kinase activity, it suppresses downstream signaling pathways involved in endothelial cell migration, invasion, and vascular sprouting. In animal models, GNE-495 demonstrates favorable pharmacokinetic profiles and effectively attenuates retinal angiogenesis, highlighting its utility in validating MAP4K4 as a therapeutic target in vascular disorders.

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
