Biological Activity

GNE 2861 is a group II selective p21-activated kinase (PAK) inhibitor that inhibits PAK4 (IC50 7.5 nM), PAK5 (IC50 36 nM), and PAK6 (IC50 126 nM). By blocking group II PAK activity, GNE 2861 suppresses cancer cell migration and invasion. Additionally, GNE 2861 disrupts estrogen receptor alpha (ERα) signaling and transcriptional activity, restoring tamoxifen sensitivity in resistant breast cancer cells.[1][2]

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
