GNE-049 is a highly potent and selective CREB-binding protein (CBP) bromodomain inhibitor (TR-FRET IC50 1.1 nM; cellular BRET IC50 12 nM) that exhibits over 3000-fold selectivity against BRD4 (IC50 4200 nM). By disrupting the interaction between the CBP bromodomain and acetylated histones, GNE-049 impairs chromatin accessibility, suppresses MYC-driven oncogenic transcriptional programs, and inhibits cancer cell proliferation. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
