GN44028 is a potent, orally active hypoxia-inducible factor-1α (HIF-1α) inhibitor with an IC50 of 14 nM against hypoxia-induced transcriptional activity. The compound inhibits HIF-1α transcriptional activation without affecting HIF-1α mRNA expression, protein accumulation, or HIF-1α/HIF-1β heterodimerization. Downstream, GN44028 suppresses hypoxia-driven gene expression, weakens tumor-initiating capability, and enhances anticancer efficacy in colorectal cancer models.

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
