Glycerophosphoric acid is a naturally occurring endogenous metabolite and substrate analogue that targets the α-site of the tryptophan synthase bienzyme complex. Upon binding to the α-site, it strongly inhibits the reaction of indole and nucleophilic indole analogues with the α-aminoacrylate intermediate E(A-A). Blockage of this active site restricts intermediate channeling through the intersubunit tunnel, impairing bacterial amino acid biosynthesis pathways and tryptophan production. [1]
