Gimeracil is a potent inhibitor of dihydropyrimidine dehydrogenase (DPD). By inhibiting DPD, gimeracil prevents the metabolic degradation of 5-fluorouracil (5-FU), sustaining effective therapeutic levels of 5-FU to enhance antitumor activity. Furthermore, gimeracil suppresses the early steps of homologous recombination repair for DNA double-strand breaks, leading to persistent DNA damage and enhanced chemosensitivity and radiosensitivity in cancer cells.
