GEM144 is a potent and orally active dual inhibitor of DNA polymerase α (POLA1) and histone deacetylase 11 (HDAC11). Dual targeting of POLA1 and HDAC11 induces p53 acetylation and subsequent p21 activation, leading to G1/S cell cycle arrest and apoptosis, which results in potent antiproliferative activity in vitro and antitumor efficacy in vivo.
