Gefapixant is an orally active small-molecule antagonist targeting human P2X3 receptors (IC, as well as recombinant hP2X3 homotrimeric and hP2X2/3 heterotrimeric receptors. By selectively blocking P2X3-containing receptors on primary sensory vagal afferent nerves, gefapixant inhibits extracellular ATP-mediated signaling. This blockade suppresses hyperexcitability of the cough reflex arc, resulting in reduced cough frequency and sensory hypersensitivity in chronic cough phenotypes. [1][2]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
