GDC-1971 is a potent allosteric inhibitor targeting Src homology 2 domain-containing phosphatase 2 (SHP2). By binding to an allosteric interface and stabilizing the autoinhibited enzyme conformation, GDC-1971 disrupts SOS1-mediated RAS activation and dampens oncogenic RAS/MAPK pathway signaling. Consequently, this inhibition reduces phosphorylated ERK levels and impairs cell proliferation and tumor growth across receptor tyrosine kinase-driven cancer models. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
