Biological Activity

GDC-0349 is a potent and selective ATP-competitive inhibitor of mTOR with over 790-fold selectivity against PI3Kα and a panel of other kinases. By inhibiting both mTORC1 and mTORC2 complexes, GDC-0349 blocks downstream phosphorylation of AKT and p70S6K within the PI3K/AKT/mTOR pathway, leading to suppression of cellular proliferation in human cancer cell lines. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
