GCN2-IN-1 (A-92) is an active site inhibitor of general control nonderepressible 2 kinase (GCN2, IC50 <0.3 μM). By inhibiting GCN2 autophosphorylation, it blocks downstream integrated stress response signaling, leading to reduced phosphorylation of eIF2α and decreased expression of ATF4. In cell models, GCN2-IN-1 suppresses adaptive transcriptional programs under nutrient stress, inhibits HIV-1 integrase phosphorylation, and modulates retroviral integration efficiency.[1][2]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
