GANT61 (NSC 136476) is a small-molecule inhibitor of GLI1- and GLI2-induced transcription that selectively suppresses the Hedgehog signaling pathway with an IC50 ofin GLI1-expressing HEK293T cells. By inhibiting GLI-mediated nuclear gene transactivation, GANT61 blocks downstream Hedgehog target gene expression, impairs cell proliferation, and induces apoptosis as well as protective autophagy in susceptible cell lines. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
