Lerociclib (G1T38) is a potent, selective, and orally bioavailable inhibitor of CDK4 (IC50 0.001 μM), CDK6 (IC50 0.002 μM), and CDK9 (IC50 0.028 μM). By suppressing CDK4/6 activity, it prevents retinoblastoma protein (Rb) phosphorylation and arrests cancer cells in the G1 phase of the cell cycle. This inhibition impairs tumor cell proliferation across multiple cyclin D-dependent cancer models.

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
