G-6599 is a monovalent degrader targeting the SWI/SNF chromatin remodeling proteins SMARCA2 and SMARCA4. By covalently binding a specific cysteine residue on the E3 ubiquitin ligase FBXO22, it facilitates ternary complex assembly and drives ubiquitin-proteasome-mediated degradation of SMARCA2 and SMARCA4. Targeted destruction of these SWI/SNF subunits impairs chromatin remodeling and transcriptional regulation, demonstrating selective antiproliferative potential in androgen-dependent prostate cancer and SMARCA4-mutant non-small cell lung cancer models.
